  encapsulation refers to traffic sent from the SPAN session and ingress means traffic from the PC connected to the destination port.

  If I use isl, traffic sent from the PC will be tagget with a isl header. Which VLAN will it use ?

send packets to the network that uses that VLAN. Without it you can only use
destination port for sniffing, not for "normal" use.
