No, the router will not set the DE bit with your configuration.

In which case/condition, the frame-relay interface will set packet
with DE-bit if I enable frame-relay traffic-shaping on serial
interface ?

If I have the following frame-relay class,for example, will it set
fr-de bit during congestion ?

map-class frame-relay FRTS
 frame-relay cir 128000
 frame-relay bc 16000
 frame-relay mincir 48000
 frame-relay adaptive-shaping becn
