I understand that when we use MTP we cannot use G729, is it correct and why ?

Without CUBE you will have to NAT all of your private IPs from your
phones/CUCM. Alternatively you could put a MTP in a DMZ. Both of these
solutions are far from optimal because the NAT option requires for your
firewall to keep up with SIP traffic/standards (historically terrible) or
for you to put an internal CUCM component reachable from the internet (not a
fantastic idea either). CUCM 5.1 is also not very good at SIP, so there's a
good chance you'll run into problems. Your provider will require early
offer, which will require an MTP on CUCM, which will mean all calls will be
G.711. I would confirm your MTP resources to make sure that's alright.

So yes, you really should be using a CUBE here.
