Well, there are buttom-up and top-down troubleshooting approaches.
Try standard tools, like traceroute, and see where the response grows too
large.
If it's low up to the server, then I would place sniffer there. If it's up
to some switch/router, check out that device.
Check other apps on that server, are they running ok?
