There would be little or no manual configuration necessary because PVST+ is
enabled by default (Cisco switches). For basic optimization, root bridge
placement and configuration of spanning-tree portfast on access ports would
need to be done either way. You wouldn't have to configure separate STP
instances and assign vlans to them. PVST+ is less scalable though since
your switch will run a separate STP instance for every single VLAN.
Once you have configured a significant number of vlans and you have need for
optimal configuration with load balancing vlans over trunks, then you should
consider MSTP. If load balancing vlans is absolutely necessary for you,
even with just a few vlans, this can be done with less configuration via
MSTP.
